A Simple Apparatus for Freezing-Drying Histological Specimens

چکیده
MANY of the conventional types of freezing-drying apparatus employ an oil or mercury diffusion-pump, backed by a rotary pump, to obtain the high vacua necessary to ensure that the water molecules which escape from the specimens have sufficient mean free path to enable them to reach the vapour trap. Clearly, the efficiency of the freezing-drying process might be greatly increased if the specimens could be placed inside a diffusion-pump designed to operate at the necessary low temperature. In such an apparatus the water molecules which escape from the specimens would be entrained in a stream of gas and carried out of the system. As considerations of mean free path would then become unimportant, the need for vapour traps and ultrahigh vacua would be eliminated. The first steps towards the design of an apparatus of this sort were taken independently by Treffenberg (1953) and Jensen (1954), both of whom effected dehydration of specimens by means of a stream of dry air. The devices designed by both these authors involve elaborate measures for drying the air and maintaining the necessary low temperature of the air and specimens during the drying process. The apparatus about to be described here is greatly simplified in that it uses solid carbon dioxide as the source of both dry gas and refrigeration. Any moisture which might be present in the carbon dioxide snow is effectively trapped, so that for practical purposes the gas evolved can be regarded as dry.

Influence of Freezing on Muscles of Rainbow Trout (Oncorhynchus mykiss): A Histological and Microbiological Study
Background: Freezing is a common and ancient method for preservation of foods which is applicable both under household and industrial conditions. The objective of the study was to establish histological and microbiological changes in dorsal and abdominal muscles of rainbow trout (Oncorhynchus mykiss) after freezing once and twice. Methods: Forty-five fresh rainbow trout specimens were distribu...

In this study, different methods for shell splitting and the effect of freezing prior to drying on shell splitting percentage of pistachio were investigated. A completely randomized design was used to investigate the effects of different freezing temperatures (0, -6, -12 and -18°C), different drying temperatures (80, 90 and 100°C) and different cultivars (Akbari and Kalehghouchi) on shell split...

The difficulties in sectioning frozen biological objects for electron microscopic investigations are overcome by Steere's freezing-etching method. In order to test this method and to open up a wide field of application, the new freezing-ultramicrotome has been designed. The apparatus consists of the combination of an ultramicrotome with freezing-drying and shadow-casting installations in the sa...
متن کاملSperm freeze-drying; effects on sperm parameters
Today, sperm cryopreservation is a routine practice in infertility clinics. One of the alternative methods to preserve sperm is freeze-drying. This process consists of three steps. The first step is sample freezing, during which the solvent is separated from the solvent. Water forms ice crystals and the solute is placed between the ice crystals. Freezing may be done in a separate device or in a...

Spinal ganglia of rabbits and cats were prepared for histological examination in a variety of ways. Dark staining multangular cells are present in sections after fixation with formaldehyde succeeded by (a) alcoholic dehydration; (b) sectioning with the freezing microtome. Sections treated in other ways, including freezing-drying, only have the typical small and large round cells. It is suggeste...
